Isolation, purification and properties of the hexaene macrolides candihexin I and candihexin II.

چکیده
The hexaene macrolide candihexin produced by the mutant 18A2 of Streptomyces viridoflavus IMRU 3685 has been found to be a complex of two component fractions which we have named candihexins I and II. Extraction and purification methods have been developed for preparation of pure candihexin complex. Separation of the component fractions has been achieved by solvent fractionation and partition column chromatography. Candihexins I and II differ in solubility in organic solvents but have similar UV spectra. Candihexin I is active in vitro against yeasts and filamentous fungi with minimal inhibitory concentrations of l-6;!g!ml. Candihexin II is essentially inactive.

Sequence of biosynthesis of the components of the polyene macrolides candidin and candihexin: macrolide aglycones as intracellular components.
A sequential formation of the single components of the polyene macrolide candidin complex (heptaene) has been found. In addition to the three components occurring in the candidin complex at the end of the fermentation, two other "early" all-trans heptaene components have been characterized. They exist only during the phase of active biosynthesis of candidin. Two of the components of the polyene...
متن کاملCandihexin polyene macrolide complex: physicochemical characterization and antifungal activities of the single components.
The candihexin complex produced by a natural mutant of Streptomyces viridoflavus has been resolved into eight components. At least six of them, having hexaene chromophores, can be separated directly by thin-layer chromatography. The four major components, candihexins A, B, E, and F, account for about 90% of the total complex. They have been isolated and purified to homogeneity. All the major co...
متن کاملHexaene H-85, a hexaene macrolide complex.
The hexaene macrolide antibiotics constitute the smallest group of the polyenes (12 ~ 1 5). Mediocidin, endomycin, hexaene H-85 and hexafungin each have a significant tetraene spectrum in addition to a hexaene spectrum (hexaene spectrum being predominant) and show high activity against fungi and moderate against Gram-positive bacteria. It was not possible to isolate the separate tetraene and he...
متن کاملOccurrence and Removal of Macrolides in Municipal Wastewater Treatment Plants: A Review
Introduction: Macrolides are a group of antibacterial agents. Given their clinical importance, and the consistent rise in resistance among pathogenic bacteria, macrolides have been the targets of extensive research. Materials and Methods: This review considered the number of macrolides in different wastewater and the removal of these drugs. The antibiotics were frequently detected in influents...
متن کاملOptimization of The Cell Aggregates Method for Isolation and Purification of Human Granulosa Cells from Follicular Fluid
Objective Aspirated ovarian follicular fluids (FF) contain luteal granulosa cells (LGC) and other contaminating cell types. Several strategies such as the antibody binding methods, the flask method, the cell strainer, and the positive selection of granulosa aggregates after density gradient centrifugation, were tested as LGCs purification methods. Each of these strategies has its own advantages...
